Curcumin steers THP-1 cells under LPS and mTORC1 challenges toward phenotypically resting, low cytokine-producing macrophages

The persistent activation of intestinal mechanistic target rapamycin complex 1 (mTORC1) triggered by mucosal stress has been linked to deregulation the gut immune response resulting in inflammation and cell death. present study investigated regulatory properties food-derived mTORC1 modulators, curcumin, piperine, toward polarization stimulated macrophages differentiation monocytes at two activity levels (baseline elevated). To that end, we created stable human THP-1 exhibiting normal or constitutively active mTORC1. Curcumin its combination with but not piperine alone, suppressed kinase activity, curtailed lipopolysaccharide-mediated inflammatory macrophages, repressed macrophage inhibiting signaling pathways involved M1 M2 (mTORC2 cAMP element binding protein) polarization. effects curcumin/piperine were modest overall, indicating it was curcumin modulated differentiating into acquiring a M0 phenotype characterized low cytokine output.
